Han er oprindeligt uddannet lærer, men i 1981 debuterede han med et tegneseriealbum i en serie om Jens Langkniv. De fleste af Orla Klausens fortællinger tager udgangspunkt i Danmarkshistorien, og en stor del af dem foregår i 1600-tallet. Klausen har desuden illustreret adskillige børnebøger.
