Mus og Mænd er den tragiske historie om de to venner George og Lennie, der vandrer fra ranch til ranch i Californien i håb om at finde arbejde under Den Store Depression i 1930’erne. George er lille, men intelligent, og Lennie er stor og stærk, men mere eller mindre retarderet. De håber en dag at kunne realisere deres fælles drøm om at slå sig ned og arbejde på deres eget stykke jord.
